Spatial characteristics of neutrophils and eosinophils in conducting airway mucosa of mice with induced allergic airway inflammation

چکیده
Daily inhaled antigens induce cellular immune response in the airways. In case of allergens, allergic airway inflammation is usually represented by eosinophils, however, neutrophil infiltration also observed during severe asthma. Animal models contribute to investigation mechanisms that involve switching eosinophil- or neutrophil-mediated inflammation. Data about spatial location eosinophils and neutrophils airways are necessary for both understanding drag potential estimation, not completely investigated. present study, we characterized model Aspergillus fumigatus extract-induced allowed investigating early stage development. The adequacy was confirmed according blood bronchoalveolar lavage eosinophilia. Using immunohistochemical staining conducting as a whole-mount confocal laser scanning microscopy, estimated eosinophil location: luminal side epithelium, wall submucosal compartment close smooth muscle layer. An activation detected upon significant elevation percentage compared intact mice. Simultaneously, number significantly increased At this time point, predominated lavages mucosa neutrophils. Spatial mucosal cell analysis demonstrated mostly located compartment, lesser extent wall, few were epithelium. Neutrophils mainly infiltrated while no wall. data suggests further allergen challenge, evidently but will migrate through lumen. Thus, can be expected damage epithelium proximity layer together with bronchoconstriction induction.

Effect of P2X4R on airway inflammation and airway remodeling in allergic airway challenge in mice.
P2X4 receptor (P2X4R) is the most widely expressed subtype of the P2XRs in the purinergic receptor family. Adenosine triphosphate (ATP), a ligand for this receptor, has been implicated in the pathogenesis of asthma. ATP‑P2X4R signaling is involved in pulmonary vascular remodeling, and in the proliferation and differentiation of airway and alveolar epithelial cell lines. However, the role of P2X...
متن کاملAirway eosinophils: allergic inflammation recruited professional antigen-presenting cells.
The capacity of airway eosinophils, potentially pertinent to allergic diseases of the upper and lower airways, to function as professional APCs, those specifically able to elicit responses from unprimed, Ag-naive CD4(+) T cells has been uncertain. We investigated whether airway eosinophils are capable of initiating naive T cell responses in vivo. Eosinophils, isolated free of other APCs from th...
متن کاملTanshinone IIA attenuates ovalbumin-induced airway inflammation and hyperresponsiveness in a murine model of asthma
Objective(s): Tanshinone IIA (T. IIA), one of the most pharmacologically active components extracted from Salviae miltiorrhiza, has anti-inflammatory and antioxidant features. The aim of the present study is to investigate the benefit of T. IIA on asthma using a murine model of asthma induced by ovalbumin (OVA). Materials and Methods: Male BALB/c mice were used in the present study. The mice we...
